I've noticed the usual bunch of morons [Image: tongue.png] posting crap messages as they often do in Forums...sad little people with nothing else better to do than to talk a load of garbage. Pleased to see the Administrator cleans these idiots off the site regularly.
I doubt that these twats have never even heard of PK. They probably think it stands for Posting 'Krap' as their spelling is much like a child!!

Information Forum guidelines for guests and members

I've just had cause to ban 1 user for inappropriate posts.
The object of the Forum is for users who have comments, questions or suggestions primarily for PCModfit or PK queries  in general. Myself or other members, I'm sure, are more than happy to help new, or established users of the program to conduct their PK analysis.
Please don't abuse the Forum with political, religious or ideology statements about subjects that are not related to PK as was the case for a member to be banned. Also, Russian is not an appropriate language for this Forum particularly in the present climate!!!

  NCA result using published data

Spotted this on the PCModfit Webpage...never noticed it before!! Tut tut!

It's a set of NCA data that was actually published - don't see this very often and I thought I'd run it through PCModfit and see what I got.
The result was identical to the published data...no surprises here as I've used PCModfit for several regulatory submissions (as many other users have) and never had it questioned!!!
I've attached the Excel sheet with the data and results + the publication results for anyone who's interested and wants to try it. Happy days are here again...!!!  Big Grin

The Administrator has just opened up some permissions on the Forum so that Guests can post queries/comments, by clicking 'Post Thread' and to send replies. The messages are usually read by myself and other members so any abuse or idiotic behaviour will be dealt with by banning the offenders IP address. Hopefully, this will help some users who don't want to become a member, even though it is anonymous, to ask for some assistance. There are some knowledgeable folk in this Forum who will be able to help you.

  Trying out V7.3 poly-exponentials and a 3-compt. oral model

To Graham (Author) and anyone else who is interested...
I've been tinkering around with models 1 to 6 as single and repeat doses using the latest V7.3. From my data sets, everything seems to work fine you'll be pleased to know. I thought I'd try out the 4-exponential oral model (model no. 3 as I've never used this one) and see how it compared to model no. 42 (never used this one either!). They should yield a very similar answer and bingo; they did! I've attached the data and my starting estimates etc. for each one to show how the two models compare as I can't see any 3-compartment oral data anywhere in the literature! Now there's a surprise!!!
I ended up with an identical sum of squares and overall fit. The compartment model took a tad longer (well, 1/2 second) which is not surprising as the complexity of the calculations far exceeds a that of a simple poly-exponential function.
The data were generated separately from the program and slightly rounded for ease of examination. I've attached the data with my starting parameters, doses and intervals etc. together with the results that I got. Methinks this demonstrates PCModfit has these models working just fine and yields an identical result really quickly. Happy bunny!!
I've tried similar comparisons with 1 and 2-compartment i.v. and oral profiles as well and they all match up nicely as well!
It was an interesting one for sure!

Just for information, PCModfit V7.3 with updates from previous versions is now released (runs on 32 or 64-bit PC computers). The modelling option has been modified to allow models 1 to 6 (poly-exponentials i.v. and oral) to be used in repeat dosing regimens as this was suggested by a few users. The models worked fine for single dose regimens but not coded for repeated doses with different doses and intervals. This option is now available and has been tested. If the user prefers the compartmental models (recommended) with micro-rates e.g., k12, k21 etc. these models can still be used for single and repeat dose scenarios as before. In most of the spreadsheets there is now a facility (updated in V7.3) to calculate micro-rate values from λ values and vice-versa as these can be tricky to calculate with multiple compartments.

I've been messing around and trying out PK data sets for fitting! Just in case anyone wants to try out the corrected model 2 issue...attached are some data that I used for testing it. V7.1 and earlier versions were troublesome with these sets for model 2 but ok for model 9. Now works brilliantly with both models in V7.2 (thanks Graham!!). The Excel sheet attached shows the set up that I used...

Quick update...
PCModfit V7.2 with updates from previous versions is now released (runs on 32 or 64-bit PC computers). The modelling module has been modified to show a progress bar after a fitting run is finished to let the user know how far the creation of the graphics in the spreadsheet and .png files has been completed. The fitting part is generally very fast but the data transfer from files into Excel® can take a little while and is usually slower than the actual modelling. Useful to the user when numerous profiles are run within the same batch.
An intermittent runtime error was found for modelling data using model no. 2 (3-exponential oral). This is now fixed and updated in V7.2.
The Superposition module in V7.2 is now at least twice as fast when compared to previous versions and yields the same results as V7.1 (tested with several different regimens). This can be very useful for longer profile times with numerous doses. The increase in speed is primarily due to modifying Font changes in the spreadsheet as the results are imported into Excel®.
Time above module has been modified to add a Profile reference to each result and Graph labels (axes, legends and title for completeness). The number of profiles maximum is 100 and each profile can now have up to 1000 data points (100 previously).

To register...
Remember, joining the Forum is free and will allow you to send messages and ask/answer questions to either the Forum or to other members specifically.

Instructions...
Click Register (just below picture at top of window).
Accept the conditions.
On the registration page enter the following.
Username: What name you would like to use in the Forum (will be seen by others).
Password: This is only for Login purposes and is Private to you only.
E-mail: Enter your normal e-mail address which is hidden if you choose (recommended).
On the right of the Registration page, for security and anonymity, the Administrator suggests you tick the box which says...hide email from other users (this prevents your email address being made public).
Any messages/questions you send will only have your Username.
Enter the text for the Image Verification and answer the Security Question.
Submit the Registration and you'll be a Member.
